The list for reference entries is extended with 145 lines for MALDI reference spectra from EMALiMB 3.0 for fungi from (RIKEN, Japan) in the MALDI-UP catalog.
Peter Kuhnert from University of Bern, Switzerland, is sharing information to msp of Reella acinonychis gen. nov., sp. nov., a Pasteurellaceae described recently.
Furthermore, several new reference entries from the meat and the plant database from CVUA Stuttgart are listed.

We have now included further MSPs for fungi from the EMALiMB 3.0 database (RIKEN, Japan) in the MALDI-UP catalog. Initial references for Mucoromycota and Mortierellomycota were also listed.
Among the parasites from EMALiMB, first msp are listed for Trypanosoma cruzi, the causative agent of Chagas disease, and for Trichomonas vaginalis.
Furthermore 14 new reference entries from the meat (3), and from the plant database (4) are included.

Response to call #02 for single spectra of Staphylococcus epidermidis with checked species status

Staphylococcus epidermidis on species level – Validation report for MALDI Biotyper shared:
On 15th January, our group made a call for single spectra from confirmed Staphylococcus epidermidis of other institutions. We got support of several MALDI users (e.g. University Göttingen, Germany; LALLF, Mecklenburg-Vorpommern, Germany), with spectra from well described isolates (e.g.from Nißler et al., 2020¸ Nature Communications 11: 5995). These single spectra are now listed in the MALDI-UP catalogue. Thanks a lot for your help!

This validation study for S. epidermidis is performed analogue the criteria from the guidelines (BVL, 2022).
Now, the report for a single laboratory is available at the tools section.

We have now included all MSPs for bacteria from the EMALiMB 3.0 database in the MALDI-UP catalog. In addition to the reference spectra from the JCM collection, the GTC isolate pool is now also integrated. The specific naming conventions of these MSPs require some practice when reading reports. We have also noted in the MALDI-UP catalog any typos or taxonomy errors we noticed of the freely available, well-curated Japanese database.
Furthermore, 14 new reference entries from the microbe (4), the meat (2), and from the plant database (8) are listed.

Our goal:
Validation of the actual database, on species level according to the Guidelines for validating using MALDI-TOF-MS (BVL) [see Tools/Validation].

FT-IR

As a new component, we have added a list of infrared spectra of microorganisms to the catalog (NEW: IR-Spectra tab). This list now contains data for 631 IR-Biotyper spectra from 91 isolates from the validation study by Oberreuter et al., 2023 for identification of serogroups of Listeria monocytogenes.
The reports of this study are presented within our FTIR section.

In der oben genannten Neuerscheinung werden für die mikrobiologische Wasseruntersuchung spektrometrische und spektroskopische Verfahren, wie MALDI-TOF MS und FT-IR, ausdrücklich eingeschlossen.
In den Anhängen werden mehrere Beispiele gegeben, wie Parameter für die MALDI-TOF MS auch im Einzellabor validiert werden können, um diese bei der Wassermikrobiologie einzusetzen (z. B. Anhang C, E, F).

This update includes a first selection of new reference entries for clinical microbes from ANLIS-Malbrán and its National Mass Spectrometry Network (ReNaEM, Argentina), compatible with the Bruker Biotyper system.
MALDI spectra from 50 institutions are now represented in MALDI-UP.
Furthermore, new reference entries from the meat database, made in a LCiP project, and a first new set of spectra for plants, made in a research semester of a student from the Esslingen university have been added.

In the Tools section, we add new Validation Reports for the Corynebacterium diphtheriae species complex, and for Listeria ivanovii, the second pathogenic Listeria spec.

Furthermore, new reference entries from the meat database made in context of a LCiP-project at the CVUA Stuttgart have been added. This will continue until the next update.

New tap “FT-IR” on MALDI-UP

Under the FT-IR tab on the MALDI-UP website, we compile tools and resources for microbiological applications of Fourier transform infrared spectroscopy.

FT-IR has been used for the characterization of microorganisms for > 25 years. The applications include identifications at the species level and determination of sero- and biotypes or pathogenicity factors. The direct comparison of spectra also often enables a rapid analysis of contamination routes.

In microbiological analytical workflows, FT-IR can be used downstream of species determination by MALDI-TOF MS.

We have posted validation reports for serotyping of Listeria monocytogenes, and Salmonella serogroups here. You will also find references to journal articles, preprints and posters.

As with the main topic MALDI-TOF MS, we would like to encourage interested readers to contribute links and hints to own work. These may also include posters or publications from the past, for example.

MALDI-UP – Status 18.12.2024

This update includes several new reference entries from the German Consiliary-Laboratory for Vibrio in food (BfR, Berlin) listed in the MALDI UP catalogue. First free available MSPs for Corynebacterium ramonii, a member of the zoonotic C. diphtheriae species complex made by CVUA Stuttgart were added.

Furthermore, the plant database has been expanded by 18 new reference entries for various berries from LGL Oberschleißheim and CVUA Stuttgart.

With this update, more than 6000 individual spectra of bacteria are listed for validation purposes.

Corynebacterium diphtheriae group – New validation reports available

Corynebacteria, which are important pathogen agents for humans and animals, are summarized in the Corynebacterium (C.) diphtheriae group. This group includes in particular C. diphtheriae (pathogen of classical diphtheria), C. belfantii, C. ulcerans and C. pseudotuberculosis. Since 2020, further species have been described within the group: C. ramonii, C. rouxii, and C. silvaticum. These species are closely related to each other, making reliable species identification difficult. To date, these important new pathogens have not been integrated in common commercial databases. User made MSPs of all members of the C. diphtheriae group are available for exchange in the MALDI-UP catalog now.

Using these new reference entries, the species identification of the C. diphtheriae group in the Bruker Biotyper system can be improved. As evidence, four formal validations from the CVUA Stuttgart for C. diphtheriae, C. ulcerans+ramonii, C. pseudotuberculosis and C. silvaticum are now presented in the tools-section of the MALDI-TOF MS User Platform.

Streptococcus equi:

Subspecies assignments of S. equi equi and of S. equi zooepidemicus/ruminatorum using the collection of MSPs and validation spectra from MALDI-UP.

Strangles, a relevant disease of horses, is connected to the subspecies equi of S. equi.
In diagnostics, the differerentiation of. S. equi equi from the similar ssp S. equi zooepidemicus is a need as ssp. zooepidemicus is frequently isolated as commensals also from horses and other sources.

The reliable differentiation of the ssp. S. e. equi is therefore a prerequisite for targeted veterinary measures.

Following the Validation Guidelines for MALDI-TOF MS Identifications in a single laboratory (BVL, 2022), reports for the subspecies differentiation via MALDI-TOF MS for these two groups were compiled. We share this reports on MALDI-UP in the tools section to activate further exchange among MALDI-UP users.

Response to call #01

Streptococcus equi on species level - Validation report for MALDI Biotyper shared:

The validation study is performed analogue the criteria from the guidelines (BVL, 2022). The report for a single laboratory version v01 is available at the tools section [see Tools/Validation].

On 5th July, our group made a first call for single spectra from confirmed Streptococcus strains of other institutions. We got support of four MALDI users (STUA Aulendorf, Germany; AGES LINZ, Austria; LALLF, Mecklenburg-Vorpommern, Germany; RWTH Aachen, Germany). Especially helpful were the spectra of the comprehensive reference strain collection of the RWTH Aachen. This support also provided evidence of the robustness of the targeted identification.

We are now continuing our effort towards the subspecies. Independently, further veterinary relevant examples of relevant Streptococcus will follow.

Any further contributions from the user community are welcome via MALDI-UP.
